 Lab Processing

  1. Clean, dry, no UV light. *
  2. Lab tech clean – wash hands – lab coat on *.
  3. Supplies in plastic containers *
  4. Good H2O(Distilled water)for making extender ( recommended is Baxter bottled H20) *
  5. Make up extender properly – temp – extender made up at least 1 hour prior to use (for it to balance and balance – proper warmer. * )
  6. Cups warmed & kept clean *
  7. Proper recording of - boars - collected every 7 to 10 days – proper semen extending records – morphology records done on each boar every month. *
  8. Is semen extended within 3 to 5 min after brought to lab?  *
  9. Calibration of sperm Q done regularly and recorded. * DAILY
  10. Check microcruvetts to make sure they read 0 - *
  11. Semen extended properly *

        Swirl container before taking samples and temp check *

        Using clean pipette for each sample *

        Warm cover slides, sample covered immediately, looked at promptly. *

        Temp. check of semen and extender each time *

        Extender into semen slowly poured up against side of container. *

        Semen checked for motility again after extending *

        Dispensing tube applied properly * USING BAG

  1. Packaging of semen

       Mix semen before packaging *

      Pour into side of tubes reducing foam *

      Mix semen after pouring every 6 to 10 does *

      Fill tubes to 80ml in proper tube color for genetic line *

      Seal promptly * YES THEY MAKE SURE CAPS ARE TIGHT.

  1. Labeling done properly  - Legible  - ID – Date
  2. All semen out of UV light * YES
  3. Storage cooling unit -  does it have a check list for temperatures – is it clean –  Does anyone look at this list to ensure compliance?  *
  4. Are doses cooled promptly that are not being used that day? *
  5. Semen not stacked when cooling – cool to 17C *
